Small Size Genomic DNA
PCR-based methods for the analysis of genomic DNA are becoming increasingly common both in research and for routine purposes. Genomic DNA on an average are between 50 - 150 kb, when the large molecule genomic DNA is used as PCR template, it might not be distributed evenly in solution when a master mix need to be made for multiple PCR reactions. High Throughput Digital PCR is a newly developed technology for high throughput real time PCR reaction, making master mix is inevitable in this technology. It is very crucial to make every molecule evenly distributed into each capilary chamber on the dynamic arrays. Large size genomic DNA molecules will result in uneven distribution while smaller size genomic will be even and randomly distributed. Quality Controls
¡ñ The quality and purity of genomic DNA were tested by spectrophotometer and electrophoresis.
A260/280 is between 1.8 and 2.0 (detected in 10 mM Tris-Cl, pH 7.5).
¡ñ Plant genomic DNA concentration is determined by pico green measurement while other genomic
•DNA concentration is determined by UV260 measurement.
¡ñ RNase treatment to ensure the RNA contamination is eliminated.
¡ñ Hind III digestion test: genomic DNA was successfully digested by Hind III.
¡ñ β-Actin expression was tested by PCR amplification for Genomic DNA or PCR ready genomic DNA.
